{"id":213,"date":"2020-06-12T01:50:15","date_gmt":"2020-06-12T01:50:15","guid":{"rendered":"http:\/\/bengalnewstimes.com\/could-a-probiotic-prevent-or-reverse-parkinsons\/"},"modified":"2020-06-12T01:50:15","modified_gmt":"2020-06-12T01:50:15","slug":"could-a-probiotic-prevent-or-reverse-parkinsons","status":"publish","type":"post","link":"https:\/\/bengalnewstimes.com\/?p=213","title":{"rendered":"Could a probiotic prevent or reverse Parkinson&#8217;s?"},"content":{"rendered":"<p><\/p>\n<div id=\"\">\n<div class=\"css-1u22pos\"><span style=\"font-size:0\"\/><\/p>\n<div>\n<p class=\"css-dmtxcr\">A new study using a roundworm model of Parkinson\u2019s disease found that a probiotic bacterium could prevent, and in some cases reverse, toxic protein buildup.<\/p>\n<\/div>\n<p>Misfolded alpha-synuclein proteins in the brain are the hallmark sign of <a href=\"https:\/\/www.medicalnewstoday.com\/info\/parkinsons-disease\/\" title=\"What is Parkinson's Disease?\" class=\"content-link css-29oowu keywords\">Parkinson\u2019s disease<\/a>.<\/p>\n<p>Many experts believe that these toxic protein clumps lead to the progressive loss of brain cells that control movement.<\/p>\n<p>But the science is not <a href=\"https:\/\/www.medicalnewstoday.com\/articles\/327470.php#1\" class=\"content-link css-29oowu\">clear-cut<\/a>, and the underlying mechanisms that cause Parkinson\u2019s remain elusive.<\/p>\n<p>Without an effective way of preventing or curing Parkinson\u2019s, treatment primarily focuses on alleviating symptoms.<\/p>\n<p>A recent line of <a href=\"https:\/\/www.medicalnewstoday.com\/articles\/324402.php#5\" class=\"content-link css-29oowu\">research<\/a> has been looking into a possible link to the gut microbiome, the trillions of microbial species that populate our intestines.<\/p>\n<p>Could changing a person\u2019s gut microbiome be a way of modifying their risk of developing Parkinson\u2019s or even serve as an effective treatment?<\/p>\n<p>A group of scientists from the Universities of Edinburgh and Dundee, both in the United Kingdom, set out to investigate.<\/p>\n<p>Maria Doitsidou, a fellow at the University of Edinburgh\u2019s Centre for Discovery Brain Sciences, is the senior study author, and the team\u2019s research features in the journal <em><a href=\"https:\/\/dx.doi.org\/10.1016\/j.celrep.2019.12.078\" target=\"_blank\" rel=\"noopener noreferrer\" class=\"content-link css-29oowu\">Cell Reports<\/a><\/em>.<\/p>\n<\/div>\n<div class=\"css-0\"><span style=\"font-size:0\"\/><\/p>\n<p>For their study, Doitsidou and her colleagues used a nematode worm model that scientists had genetically engineered to express a human version of the alpha-synuclein protein.<\/p>\n<p>These worms normally develop aggregates, or clumps, of alpha-synuclein at day 1 of their adulthood, which is 72 hours after they hatch.<\/p>\n<p>However, when the researchers fed worms a diet containing a probiotic bacterial strain called <em>Bacillus subtilis <\/em>PXN21, they observed \u201ca nearly complete absence of aggregates,\u201d as they state in their paper. The worms still produced the alpha-synuclein protein, but it did not aggregate in the same way.<\/p>\n<p>In worms that had already developed protein aggregates, switching their diet to <em>B. subtilis<\/em> cleared the aggregates from the affected cells.<\/p>\n<p>The team then followed a set of worms through their lifespan and compared a <em>B. subtilis<\/em> diet with a conventional laboratory diet.<\/p>\n<p>\u201cThe maximum number of aggregates reached in animals fed with <em>B. subtilis<\/em> was far lower than that observed on the [standard] diet, indicating that <em>B. subtilis<\/em> does not simply delay aggregate formation,\u201d the authors explain in the paper.<\/p>\n<p>\u201c<em>B. subtilis<\/em> PXN21 inhibits and reverses [alpha-synuclein] aggregation in a [roundworm] model,\u201d they note.<\/p>\n<p>Is this effect specific for <em>B. subtilis<\/em> PXN21, though? To answer this question, the team compared a number of different strains of the bacterium and found that they had similar effects.<\/p>\n<\/div>\n<p><span class=\"css-zya54r\"\/><\/p>\n<div class=\"css-0\"><span style=\"font-size:0\"\/><\/p>\n<p>To find out how <em>B. subtilis<\/em> is able to prevent and clear alpha-synuclein aggregates, the team used RNA sequencing analysis to compare the gene expression of animals receiving a standard diet with that of those receiving the probiotic.<\/p>\n<p>This analysis revealed changes in sphingolipid metabolism. Sphingolipids are a type of fat molecule, and they are important components of the structure of our cell membranes.<\/p>\n<p>\u201cPrevious studies suggest that an imbalance of lipids, including ceramides and sphingolipid intermediates, may contribute to the pathology of [Parkinson\u2019s disease],\u201d the authors comment in the paper.<\/p>\n<p>Yet, changes in sphingolipid metabolism were not the only pathways that the researchers identified.<\/p>\n<p>They also saw that <em>B. subtilis<\/em> was able to protect older animals from alpha-synuclein aggregation through both the formation of complex structures called biofilms and the production of nitric oxide. In addition, the team saw changes in the dietary restriction and the insulin-like signaling pathways.<\/p>\n<p>Importantly, when the team switched animals that had first received a standard diet over to a <em>B. subtilis<\/em> diet, their motor skills improved.<\/p>\n<blockquote class=\"css-34gtoi\">\n<p><span class=\"pullquote\">\u201c<\/span>The results provide an opportunity to investigate how changing the bacteria that make up our gut microbiome affects Parkinson\u2019s.
